In vitro fertilisation laboratories reproduce biological processes that normally occur inside the woman’s body. It is therefore important to have controlled and stabilised parameters such as light, temperature, oxygen, CO2, humidity, volatile particles and pollutants in the environment, and to be able to simulate them as closely as possible to in vivo conditions.
To reduce the danger of possible contamination in the laboratory, the laboratory should be located in a non-transit area and preferably in an operating theatre area, where aseptic conditions are guaranteed. In addition, the laboratory should be equipped with a continuously operating air purification system to remove airborne particles and micro-organisms.
When handling gametes and embryos, always work in a laminar flow hood, which provides a sterile environment to avoid contamination of the culture media.
Light is an important parameter to control. Low light intensities must be used, both in the microscopes and magnifying glasses under which gametes and embryos are observed and in the laboratory itself.
Gametes and embryos are very sensitive to temperature changes. Therefore, they should be out of the incubators as little time as possible and when they are being handled, all working surfaces should be heated (laminar flow hood, microscopes, etc.). To check that the temperature is constant and adequate, the apparatus should be checked daily for proper functioning.
Gamete and embryo culture is carried out inside . Nowadays, these devices are equipped with a system to regulate the internal temperature, humidity and CO2 concentration. In this way, in vivo conditions can be simulated more accurately, which improves embryo quality and increases success rates.
